In DeSu, Cain had died to allow for reincarnation but since he is still alive in Supernatural he is instead the ancestor to Amici. This allows for their own version of the Cain and Abel tale to take place during Belial’s kidnapping of Joyce.
Joyce and Amici are cousins instead of siblings. Their mothers were twin sisters but Amici was born while Joyce’s mother was on the run.
